What is it?
Gallium Maltolate is an orally-active formulation that is a potent blocker of ribonucleotide reductase, an enzyme that promotes tumor growth. Gallium is also known to concentrate in malignant tumors and sites of infection and appears to favorably impact calcium deposition, making bones more resistant to degradation caused by cancer metastasis.

Gallium maltolate is known to retard the spread of malignant cells primarily due to its ability to mimic ferric iron (Fe3+). Ferric iron is essential for DNA synthesis, as it is present in the active site of the enzyme ribonucleotide reductase, which catalyzes the conversion of ribonucleotides to the deoxyribonucleotides required for DNA. Gallium is taken up by the rapidly proliferating cancer cells, so the cells cannot reproduce and they ultimately die by apoptosis (the process of programmed cell death).
Upon administration of Gallium Maltolate, which is structurally similar to the ferric iron (Fe3+), competes with and replaces ferric iron Fe3+. In rapidly proliferating cells, such as cancer cells, high amounts of iron are needed for DNA synthesis. The incorporation of Gallium Maltolate inactivates the ferric iron Fe3+ dependent enzyme ribonucleotide reductase (RR), an enzyme essential for DNA synthesis, leading to blockage of DNA synthesis thereby enabling cell death in rapidly proliferating cells.
Gallium Maltolate similarly reduces bacterial cell growth. In addition, Gallium Maltolate is able to suppress inflammation through the down-regulation of pro-inflammatory cells and the inhibition of pro-inflammatory cytokine secretion. Gallium Maltolate also exerts analgesic effects due to the blockage of ferric iron Fe3+-dependent enzymes involved in inflammation and may interfere with the activity of certain metalloproteinases and neuropeptides that are implicated in pain. Also, Gallium Maltolate is able to block bone resorption by osteoclasts, may block metastasis to bone, and may prevent the destruction of bone by tumors.
